Title: Targeting multiprotein complexes with small molecules

Abstract

Multiprotein complexes regulate many fundamental processes in living cells, including transcription, splicing, translation, protein folding/degradation and signaling. These multiprotein complexes are typically composed of at least one enzyme and a number of non-enzymatic cofactors. These cofactors associate either stably or transiently with the enzyme to regulate its activity, subcellular location and/or selectivity. In addition to their important physiological functions. these multiprotein complexes are also involved in the pathogenesis of human diseases. Development of novel small molecules to reverse the aberrant role of the protein complexes can be developed into therapeutics. There are tremendous opportunities in modulating the function of multiprotein complexes, including by binding to the enzyme's active site, allosteric site or protein-protein interaction interface. In this talk, I will show several projects using different approaches to target multiprotein complexes.

Dr. Hao Shao received doctoral degree in 2014 from the University of Nottingham under the supervision of Prof. Peter Fischer and Shudong Wang, he then did postdoctoral training with Prof. Jason Gestwicki at the University of California, San Francisco. In 2021. he started as assistant professor at Hunan Key Laboratory of Molecular Precision Medicine, Xiangya Hospital, Central South University. Dr, Shao's research centers on the discovery of novel small molecule drugs to treat cancer and neurogenerative diseases using multidisciplinary approaches, including medicinal chemistry, chemical biology and pharmacology. He is actively working on small molecule inhibitors of cyclin-dependent kinases and molecular chaperones, such as heat shock protein 70 and 60.
